Understanding the physiological function of PANcreatic DERived factor:  is it involved in diabetes?

PANcreatic DERived factor (PANDER) is a recently cloned cytokine-like protein with 235 amino acids that is predominantly expressed in pancreatic islet. Previous study indicated that PANDER is capable of inducing apoptosis of human and rodent islet cells. My postdoctoral research work in the past three years was focused on:

1)   Structure-function studies of PANDER. Demonstration of PANDER’s structure-

function relationship will be useful for designing and developing its agonists or  antagonists if possible in the future.

2)  Secretory mechanism of PANDER in pancreatic beta cells. PANDER is the first   

cytokine had ever been demonstrated to be co-secreted with insulin through the same regulatory mechanism following glucose stimulation.

3)  Identification of the potential target tissue(s) for PANDER and demonstration of its 

physiological function. Liver is a novel target tissue for PANDER secreted from pancreatic beta cells; PANDER inhibits liver cell insulin signaling in vivo and in vitro.

 

Our studies indicated that PANDER might be a novel cytokine playing important role in insulin resistance and type 2 diabetes. Our final goal is to set PANDER as a biomarker for diagnosis of diabetes, and to develop some agonists or antagonists for prevention and treatment of diabetes.
